The rooted tissue culture seedlings are directly transplanted in the natural environment from the test tube, and it is often difficult to survive because the external environmental conditions change too much. For this reason, it is necessary to improve the ability of tissue culture seedlings to adapt to changing external conditions by domestication before transplanting. The method is to take the tissue cultured seedlings out of the culture bottle, wash off the culture medium with tap water, plant them in a nutrient bowl filled with dried and sterilized culture soil (vermiculite), place the nutrient bowl in a manual control room, and keep it indoors. The relative humidity is above 70%, and the temperature is controlled at 16 to 20 degrees Celsius. After the domesticated tissue culture seedlings are transplanted to the greenhouse for cultivation for a period of time, they are then transplanted to the open field for cultivation.
